This product is used to semi-quantitatively detect the level of luteinizing hormone (LH) in women's urine in vitro, to predict the time of ovulation, and to guide women of childbearing age to choose the best time to conceive or to guide contraception during a safe period.

Luteinizing hormone (LH) is a glycoprotein hormone of gonadotropin, a macromolecular glycoprotein secreted by basophils in the anterior pituitary gland formed by the non-conjugated combination of α and β subunits. The β subunit has configurational features that determine its molecular specificity, which can identify appropriate target tissues, and bind to specific receptors, thereby exerting its physiological function, and its main role is to stimulate the release of mature eggs in the ovary. Normal women maintain a small amount of luteinizing hormone in their body, and the secretion of luteinizing hormone increases rapidly in the middle of menstruation, forming a "luteinizing hormone peak". The peak of luteinizing hormone release is closely related to ovarian ovulation. By continuously monitoring the concentration of luteinizing hormone in urine samples, this kit can know the changes of luteinizing hormone in female urine. Once the peak of luteinizing hormone appears, it indicates that the ovaries are about to ovulate within 24 to 60 hours.

At present, the commonly used luteinizing hormone (LH) detection methods in clinical or laboratory include chemiluminescence, immunochromatography and so on.

[Testing principle]

The product adopts the principle of double antibody sandwich. When luteinizing hormone is present in the sample, the LH antigen in the sample reacts with the colloidal gold-labeled murine antibody (LH mAb1) on the binding pad to form a labeled antibody-antigen complex. The complex is chromatographed upward by capillary action and captured by the detection line (T line) murine antibody (LH mAb2) coated on a nitrocellulose membrane, and a red band appears. The complex continues to be chromatographed upwards and is captured by a control line (C line) antibody (goat anti-chicken IgY polyclonal antibody) coated on a nitrocellulose membrane, and a red band appears. This is the criterion for judging whether the sample size is adequate and whether the chromatographic process is normal, and also serves as an internal control for this kit.

The semi-quantitative reagent mainly compares the actual chromaticity of the T line with the standard colourimetric card provided with the kit, records the corresponding chromaticity value, draws the luteinizing hormone curve, and obtains the subject's " Luteinizing hormone peak" value.

Operation Flow & Result Interpretation

Observe and record the test results of LH detection

(1) Both the detection line (T line) and the quality control line (C line) have red bands. Compare the color of the test line (T line) with that of the LH colourimetric card, find out the color scale of the color card that is closest to the color of the T line, and record the value (measurement result) and test time.

(2) There is no red band on the detection line (T line), but there is a red band on the quality control line (C line), that is, the LH content in the urine sample is 0, and the test result is recorded.

(3) There is no red band in the quality control line (C line), indicating that the test result is invalid. In this case, read the instructions carefully again, and retest with new kits. If the problem persists, stop using this batch of products immediately and contact the local supplier or manufacturer.
